A Study on the preservation of microorganisms

H. influenzae is one of the most fastidious organism in growth. Ordinary freezing stock method keeps bacteria vitality in different period. Pneumococcus maintains about 8 weeks.
Beta-haemolytic Streptococcus several months and Staphylococcus & some strain of Enterococcus keep over one year to a few years. However, H. influenzae only keeps a few weeks by the ordinary -20℃ freezing preservation method. Author intends to prolong the vitality of H. influenzaw and deviced following method.
Basic stock culture broth conists of nutrient broth 8.5mL and glycerine 1.5 mL ratio.
The yeast hydrolysate is produced 20g of commercial dry yeast with 100mL of distilled water which treated with freezing and thawing three times, then centrifuged.
After centrifugation the supernatant filtrates with Seitz filter.
The methods of preservation for H. influenzae were observed which may be applicable in small laboratories.
Compardd with regard to its simplicity, inexpensive and also easily available materials in any locality were discussed.
The most recommendable procedure is the use of special preservation media to which 10% yeast hydrolysate was added.
